2． Anhydrous sealing agent ： Such as neutral gum , Malaysian resin and artificial resin . For paraffin , Sealing of frozen and collodion slices , When used, the tissue sections must be dehydrated , Transparent and thorough , Otherwise, there will be cloud like turbidity in the film , Hinder microscopic examination .
 Usually these sealants are a kind of resin （ Natural or synthetic ） Dissolve in its natural solvent or xylene until it is in a suitable fluid that is easy to dry and solidify for sealing . Its viscosity is good for easy elimination of bubbles and free flow between cover glass and slices , A little thicker can avoid the formation of air gap under the cover glass after drying .

 Such as ： Dyeing with alkaline aniline dyes requires sealing in an acid free sealing agent . Slices like Prussian blue reaction should be sealed in a non reducing sealing agent .

 Undyed tissue is best sealed with a sealant with a high or low refractive index . While the stained sections have a refractive index of 1．52 to 1．54 The sealing agent between is the most transparent .
 Our commonly used anhydrous sealant is the commercially available neutral xylene gum , Also known as neutral gum ; It is a colorless or yellowish transparent liquid , Because acid or alkaline sealants can cause discoloration of tissue sections , Therefore, neutral sealing agent should be used . The sealing agent should be stored in a colored bottle to avoid sunlight , And stored in the dark to prevent acid change .

The only disadvantage is that with the extension of storage time , Gum gradually oxidizes xylene , The production of toluene acid and phthalic acid can make basic dyes fade .
